RNA Seq in Filamentous Fungi
RNA sequencing has become the gold standard for genome wide profiling of differentially expressed genes. However, RNA quality is crucial for any RNA Seq project and in particular fungal species are known to have high contents of polysaccharides impairing the RNA quality and quantity.
The goal of the current project was to study the differential gene expression during the interaction of closely related or and even sibling species of Ascomycetes. Beyond the RNA sequencing, Microsynth was challenged by developing a solid RNA isolation protocol as well as to implement stringent quality assurance measures before the next-generation sequencing to assure the identity/purity of all input samples.
